Webinar recording: A lysis-free approach to measuring RNA encapsulation efficiency

Encapsulation efficiency is one of the most widely measured attributes for characterizing RNA-LNP formulations. It is also one of the most labor-intensive and variable to obtain.

Conventional methods require detergent-mediated LNP lysis and around 8–10 sample-preparation steps, creating multiple opportunities for operator error and inconsistent results.

In this webinar, Dr Aida López Espinar explained how Scatter-Free Absorption Spectroscopy (SFAS) enables RNA encapsulation efficiency to be measured without lysing the LNP sample.

Watch to learn:

  • Limitations of the traditional RiboGreen assay, including partial nanoparticle disruption and Triton variability
  • How CloudSpec's integrating sphere technology captures scattered light to measure true RNA absorbance at 260 nm — without lysing your nanoparticles
  • A novel method for determining free RNA concentration using RiboGreen fluorescence detected as negative absorbance at 560 nm
  • The one-shot approach: measuring both total and free RNA from a single sample (optimized for non-absorbing lipids like SM102)
  • Cost savings of ~40% compared to the regular RiboGreen assay with no standard curves, no Triton, no black 96-well plates
